The India Ink Preparation Test is a simple microscopic examination used to detect encapsulated fungal organisms, most commonly Cryptococcus neoformans, in clinical samples such as cerebrospinal fluid (CSF), sputum, or other body fluids.
In this test, India ink is used as a negative stain, which creates a dark background while the capsule of the organism appears as a clear halo around the yeast cells. This makes it easier to visually identify the presence of encapsulated fungi under a microscope.
