The test preparation involves collecting early morning deep-cough sputum samples over 2–3 consecutive days to maximize detection of TB-causing bacteria. No specific fasting is required for sputum, but patients should rinse their mouths with water, avoid food 1-2 hours prior, and avoid smoking for 24 hours.
